Hmmm...I know certain distributors prefer a specific aspect ratio...but I'm not sure if it would 'affect' distribution. I'd say shoot in 16:9 and you'll be fine. What's the aspect ratio you're going to use?
Shoot anamorphic it shouldn't effect distribution for DVD you can pull it down to 16x9 or 4x3. If it's theatrical call the lab they can tell you the in's and out's of doing a print and what ratios work. Personally I would be concerned with getting the best actors I could afford and my story was tight and marketable, and good sound. You can have a film that looks beautiful but if the story is weak and actors aren't on and the sound sucks it won't get far. On the other hand if your actors are great the story is tight and compelling and you can hear every word of dialog but the look is not as good I think it's more forgiving.
Also now days more that ever (at least for indies) genre is a factor. If you're doing a romantic comedy or straight up drama you need A-list actors to sell it. Even some genre is getting oversaturated with product Horror for instance is an example of a genre that now you can't hardly make your budget back because everyone is doing one. Distribution has a learning curve hopefully we learn from others on what mistakes not to make. I think aspect ratios are only one very small piece of the game that is selling our film.
