Does inappropriate or mature content give you less of a chance to be selected, or be the basis of not being screened? I'm talking scenes of rape, violence, disturbing content, swearing, ect..

That is true, especially if you want to get past a jaded film festival screener. But some things, like rape and suicide will backfire IMO.
All I can say is good luck! (And suggest you do test screenings with strangers.)
If it's a short, say like 10 minutes, then there might not be enough time to develop characters before the rape. You might have to just jump into maybe. Not sure if that's still good or not.
